It depends on what you want to do with the shot.
Just about anything from 15-40 grains will work in it but your best accuracy will probably be around 25 grains of 3Fg black powder.
There is a general guideline that says the most efficient large powder load is equal to 11.5 times the bore volume.
If your .45 has a 10 inch long barrel that would be 18.29 grains.
As I say, this is just a general guideline and I'm not sure it really applies to pistols because of the small volume of the bores.
Using more than this amount does increase velocity and the muzzle flash (and BOOM) will increase as well.
