I'm building a 40 cal 1800s flintlock and I'm wanting to turn a powder measure out of some lilac I've got. How do I figure out what the inside dimensions should be?

dimensions to suit your comfort zone!
You can adjust the delivered charge by pouring in some bee's wax and using the drill bit you bored with, in hand, remove some wax for fine tuning!

Fill your favorite powder measure with black powder (or sand if you want). Drill out your new measure until the test volume fits. Might take a few tries....
 
Try boring various size/depth holes in a scrap piece of wood and measure powder that fills them. Then adjust if need be on your intended powder measure.
