Remove one nipple and lightly grease that chamber.
Dry ball the chamber then with a soft brass or aluminum NOT STEEL!! punch knock out the ball and see what kind of seal that gives.
In my experiment the balls are hand cast in a Lee ,451 round ball mold.
My experience was sprue up, down, or sideways I got a very good seal using soft lead .451" balls in a both a Pietta and a Uberti cylinder both with lightly chamfered chamber mouths.
Try it and see what happens YMMV.

I think if the balls are tight enough not to back out

This is exactly what happens in my Ruger Old Army after I had the chambers chamfered. Anything I can do to prevent the backing out? It actually does seem to be more accurate since the chamfering!
 
Not a clue. I don't have that problem with my Uberti or Pietta guns. My chambers just have the sharp edge removed not deeply chamfered. Perhaps the problem is if you are shooting heavy loads. In my .44 I use 18 to 20 grains volume of FFFg GOEX. If you are using the "P" stuff or 7cubed that may be the problem.
If you can't get real Black Gun Powder you might try APP, Black MZ, or Jim Shokey's Gold which is the same stuff with a different label. It is a different chemistry than the "substitutes" but works well using VOLUME equivalents not weight.
Produces less fouling than real BP, does not like compression, but might be easier to find.
It does not work at all in flintlocks so I am told.
