Pedersoli uses three thread sizes
1/4x28
8x1.25
6x.75
I would try running a 1/4-28 tap down the bolster just to check before buying your nipples.

Rather than using a tap that can cut new threads, I would try a 1/4-28 bolt.

Newer Pedersoli nipple seats in rifles and shotguns are 1/4-28. The bolt is cheap and worth using for an initial trial.

Would the 1/4 x 28 size die be correct for the Pedersoli .45 cal. sidelock rifle for nipple channel repair?
That would be a tap. I have rethreaded all my long guns to 1/4-28 in the name of making life simpler. You can retap most rifle, shotgun (no hand guns or muskets) without drilling. Just start a finishing tap square and do it.
