I have a .54 caliber longrifle that was dialed in 2-inches high at 50-yards and ended up a bit over 4-inches low at 100-yards. Where I hunt there are often 100-yard shots and some a tad over. Today I said "what the hay" , and I filed the front sight until my point of impact was dead-on at 100-yards. I think it will end up around 5-inches high at 50-yards, but again this isn't intended to be a 50-yard rifle. If a critter shows up at 50-yards and I am tight into the vital pocket I believe a 5-inch high impact will still be fatal (maybe end up a high shoulder hit at most). I hope I don't regret this. I do have another front sight blank if I want to start over, and other rifles for closer range conditions. So, I have my designated "long range" rifle......
 
Spark, I use the exact same charge in my .54 Renegade. You're guessing pretty close, but I think it'll be around 3.5"-4" high at 50. At least from what I see with that charge and a pretty darn tight fitting PRB.
How you hold is dependent on your preference.
While Pete G would just use a fine bead at 50, I myself would use the same sight picture I always do and just hold on the heart. (I like the heart as table fare and so try to go over it through the lungs)
