I think you should spend several days experimenting with patch and ball thickness and then powder charges until you find the powder charge that gives you pinpoint accuracy.....
Be aware that due to harmonics, one can ALWAYS find at least 2 powder charge levels (a low to medium and a high) that give the best inherent accuracy of the barrel as fired. On longer barrels one can often find 3 different charges.

As most have said I just adjust the POA. I get the most accurate load I can for each rifle, zero at 25 yds which will put it on a no hold over or under out to 100 yds with most guns. The only issue I have with this style is very close targets under 25 yds
I just don't shoot at the range at closer ranges to learn where to sight.

This comment is my bookmark so I can peruse it at leasure. I like the possibility of calculating variables and confirming them at the range.
 
I started some experimenting today. Things are hampered by my eyesight and shortcomings as a 100 yard shooter. Going from 60 to 75 grains gave me a better group and a little more elevation. I tried 80 grains which didn't seem to give me any more elevation and went back to 75 grains which gave me a slightly better group than 80. I then shot 3 rounds at 75 grains still using a center hold but holding the front sight blade just a little higher in the rear sight notch. That gave me the elevation. I think I'll stick with using 75 grains at 100 yards and finding just the right amount that I have to raise the front blade sight for elevation. Funny how accurate that gun is with 60 grains at 50 yards and then spreads out at 100 so much. It will take a lot more shooting to see where it will settle out at 100 yards but at least I have a direction to go now.....more powder and adjust sight picture while still keeping a center POA.

On the range with my .45 cal rifles (three of them), I'll increase my charge from 55 gr. to 65 to raise the poi. I still have to hold a 12 o'clock on the black instead of center as I do at 50 yards.
