A 50 PRB will drop deer fine if you hit the boilerworks. If you sight in at 75yds, you'll be a bit low at 100 and high at 50. Will depend on the gun/load. A good load will have shots touching at 50yds. When you get an accurate load, try different distances to see where the groups end up.

Wonderwads protect patches (burn through) with PRB or the base of bullet with a conical (leading). Most have a lube on them
 
Hmmm....so I'll get the wonder wads, file my front site down a bit because my peep is maxed out on elevation and try and get 3 inches high at 25 yards. 80 Gr pyrodex P seems to be the accuracy load. Will the wads do anything to the load as far as improving or taking away accuracy?
 
Some guns, some loads, or some shooters do well with a wad under the ball. It’s real handy in smooth bores.
Ball in most ml will have a two to three inch mid range tajectoy if sighted at a hundred yards. If sighted at fifty will have around a three inch drop at a hundred.
Balls look a little puny on paper compared to modren projectile, most loads will deliver 4-600 foot pounds of energy at 100 yards. Though that sounds light it’s enough to make a big hole through deer sized targets. I’ve killed more then one deer these past forty years of shooting ball only, elk and antalope (pronghorn) . Ball has never let me down except for my first smooth bore kill. I hit liver and not chest. Heard another shot as I was following a big blood trail went about 1/4 mile, and found another hunters standing over my deer. It was killed by a close range head shot. We split the meat. There was barely any blood around that head shot, and I think the deer was dead when the other hunter walked up on him.

I wouldn't do ANYTHING to the sights with a file until you find a load that shoots a group of at least 3 rounds measuring 1 1/2" to 2" at 50 yards or better.

Where the rounds got on the target right now is not important... keep aiming at the same place, record your load and group, then raise or lower your powder charge and see what that does to your group.

Only after you find a load your happy with should you file your sights.

The wonder wad tends to reduce gas cutting of conicals, which will help accuracy.

I sight my rifle 2" high at 50 yards, dead on at 75, 4" low at 100.
 
I checked Lyman data, the 370 gr conical @80 grs fffg gives 1400fps. The data shows a 100 yard zero gives a mid range of 3.27" high @ 50 yards.
With a 100 yard zero it is down approx 6" @ 125y, 15" down @ 150y.
Velocity is 994fps @ 100y, 939 @ 125y, 892 @ 150y.

I shoot a 58 cal with a 500 gr minie, so my data is different from yours.
