Use of revolver wad

Sure you can use a wad with a conical. The gains by doing it might be debated though.

If you are talking about a revolver, the wad will take up powder space in the chamber. This coupled with the length of the conical will reduce the amount of powder you can load into the chamber so the shot will be less powerful. Now, there are a lot of people who do use wads under roundballs loaded into their revolver. They feel it helps prevent chain-fire's and lubes the bore when the shot is fired.

If we are talking about a single shot pistol, the effect on the powder load won't be a problem but I have to wonder if the extra cost of the wad would be worth it.

With a rifle, where long distances are being shot the wad can help to seal off the rear of the conical so blow by is reduced. This can make the pressure behind the slug more consistent which can lead to better accuracy. I'm not convinced this benefit would be seen at the close distances a pistol is normally shot at.

At least, this is my opinion and I think it might be worth at least a half-cent?
